Target Background

Interleukin-1β (IL1b) is a pro-inflammatory cytokine important for immune responses to infection and injury. It is produced as a 31 kDa precursor, termed pro-IL-1b which is cleaved by caspase-1-dependent processing for secretion. IL1b is expressed in a wide variety of tissues and cells, especially in macrophages in the thymus, spleen, lymph nodes, Peyer's patches and bone marrow. Disruption of IL1b signaling can lead to inflammatory disease and malignancies.

Protocol Outline
  1. Prepare all reagents, samples and standards as instructed in the manual.
  2. Add 100 µl of standard or sample to each well.
  3. Incubate 2.5 h at RT or O/N at 4°C.
  4. Add 100 µl of prepared biotin antibody to each well.
  5. Incubate 1 h at RT.
  6. Add 100 µl of prepared Streptavidin solution to each well.
  7. Incubate 45 min at RT.
  8. Add 100 µl of TMB One-Step Substrate Reagent to each well.
  9. Incubate 30 min at RT.
  10. Add 50 µl of Stop Solution to each well.
  11. Read at 450 nm immediately.

Storage/Stability

    We've been looking for an ELISA kit that detects IL-1b in tissue lysates for ages. We used this kit with mouse brain tissue homogenized in a lysis buffer containing 0.25% SDS and diluted the samples 1:5 as recommended by the manufacturer protocol and got beautiful results.

    I measured the Mouse IL-1 beta (Cat#Ray Bio Mouse IL-1 Beta ELISA KIT) in Kidney tissue homogenates from mouse model of Diabetes (DB/DB-UNX) with Uni nephrectomy (UNX). I see the increased IL-1B levels in DB/DB-UNX compared to control Non-Diabetic (DB/M) mice. In this models we knew that there is increased inflammation together with Inflammasome signalling in the kidney of DB/DB-UNX. Therefore, I am interested in measure the IL-1B in our mouse of model of Diabetic kidney disease. This Kit from RayBiotech is working great in our mouse model and I would recommend this product.
