Immunogen was polypeptide Pbtx-3. This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with the immunogen. The IgG fraction of tissue culture supernatant was purified by Protein G/A affinity chromatography.
Recommended Applications
ELISA (recommended work dilution = 1:320,000)
Reconstitution
Product is supplied as a powder obtained from lyophilization of purified antibody in glycine solution. Reconstitute the antibody with sterile water to a final concentration of 1 mg/ml.
Brevetoxin (PbTx), or brevetoxins, are a suite of cyclic polyether compounds produced naturally by a species of dinoflagellate known as Karenia brevis. Brevetoxins are neurotoxins that bind to voltage-gated sodium channels in nerve cells, leading to disruption of normal neurological processes and causing the illness clinically described as neurotoxic shellfish poisoning (NSP). PbTx-3 is a potent allosteric voltage-gated Na+ channel activator and has multiple active centers (A-ring lactone, C-42 of R side chain).PbTx-3 has a high affinity to site 5 of the voltage-sensitive Na+ channels, inhibits the inactivation of Na+ channels and prolongs the mean open time of these channels. PbTx-3 repeated exposures can lead to prolonged airway hyperresponsiveness (AHR) and lung inflammation.
