Instructions

  1. Resuspend the cell pellets in any physiological buffers at the appropriate ratio, for example, 10~20/1 (V/W, 10-20 ml buffer/1 g wet cells). Typically for bacteria, we use 20 mM Tris-HCl (pH 8), 2.5 mM MgCl2, 50 mM NaCl.
  2. Add appropriate Universal Nuclease to the suspension (normally adding 250 U enzymes/1 g wet cell paste)
  3. Break the cells through ultrasonication, tissue homogenizer, freeze-thaw cycles, high pressure homogenizer, etc.
  4. Clarify the cell lysate by high speed centrifugation at 20,000 g for 30 minutes at 4°C.
  5. Transfer the clear supernatants to new tubes for downstream work.

The product is shipped with ice packs. Upon arrival, immediately store it at the temperature recommended below. It is also supplied with one vial of 10x Cleavage Buffer (200 mM Tris-HCl, 25 mM MgCl2, 500 mM NaCl, pH 8.0), 1 mL.
Storage/Stability
Use a manual defrost freezer and avoid repeated freeze-thaw cycles. For long term storage, it is recommended to store at -20°C or -80°C in appropriate aliquots. The shelf life is up to 6 months from the date of receipt at -20°C or -80°C under sterile condition.
