Protocol Outline

Prepare all reagents, samples and standards as instructed in the manual.
Add 100 µl of standard or sample to each well.
Incubate 2.5 h at RT or O/N at 4°C.
Add 100 µl of prepared biotin antibody to each well.
Incubate 1 h at RT.
Add 100 µl of prepared Streptavidin solution to each well.
Incubate 45 min at RT.
Add 100 µl of TMB One-Step Substrate Reagent to each well.
Incubate 30 min at RT.
Add 50 µl of Stop Solution to each well.
Read at 450 nm immediately.

Storage/Stability

Used it for cell culture media from prostate epithelial cells and fibroblasts. Very easy to use kit and protocol. Unfortunately, samples were significantly above standard so will have to dilute and try again. Overall good product though.

We used this kit to measure the HGF release from the tissue scaffold. It is easy to use and give a reliable result. However, we did have to try a couple of different dilutions of the samples before our release is within the range of the test capacity.

The product is really useful for detecting HGF levels in serum of breast cancer patients by just following the instructions provided in the ELISA kit. We observed HGF levels are high in aggressive breast cancer compared to less aggressive tumor. We like it very much.

This product is easy to use. The protocol is also very straightforward. I just need to follow the manufacturer's instructions and data is very clean and beautiful. Only shortcoming is that is not very sensitive and I cannot dilute serum sample too much. It's possible that the serum levels of HGF is lower than other secreted protein in human serum. Overall is good!
