Target Background

Platelets in blood are connected to D subunits. When blood clots form, the D subunits between two platelets forms a bond or a D-dimer. When the blood clot is broken down by fibrinolysis the D-dimers break apart from the platelet. Therefore, the D-dimer concentration can be determined using a blood test to diagnose blood clotting disorders such as, deep vein thrombosis, pulmonary embolism and disseminated intravascular coagulation.

Protocol Outline
  1. Prepare all reagents, samples and standards as instructed in the manual.
  2. Add 100 µl of standard or sample to each well.
  3. Incubate 2.5 h at RT or O/N at 4°C.
  4. Add 100 µl of prepared biotin antibody to each well.
  5. Incubate 1 h at RT.
  6. Add 100 µl of prepared Streptavidin solution to each well.
  7. Incubate 45 min at RT.
  8. Add 100 µl of TMB One-Step Substrate Reagent to each well.
  9. Incubate 30 min at RT.
  10. Add 50 µl of Stop Solution to each well.
  11. Read at 450 nm immediately.

Storage/Stability

    This kit works fine. We bought 6, and there was no issue with any. My one problem is that in the protocol, the dilution range for serum is 'between 1:5,000 and 1:500,000'. That is a very big range. So first I had to run a test run to see where in that range I should dilute my samples (it was 1:500,000 for HIV infected adult men), and then I had to do serial dilutions - 4 of them - to get up to 1:500,000. I was afraid I was losing accuracy with all that extra pipetting with multichannels. I which the kit was designed differently to address this.
